摘要
基因标记和光子学显象技术相结合,实时观测中性粒细胞吞噬病原体细胞骨架运动规律. 利用致冷CCD系统组建了高灵敏度的荧光激发与测试装置. 将带有绿色荧光蛋白(GFP)基因的PRSETB质粒转化大肠杆菌BL21,利用BL21高效表达GFP后发出强烈绿色荧光的特点,用光子显象技术连续记录荧光的变化,从而在不受干扰的条件下实时动态观察了中性粒细胞粘附、内吞、消化大肠杆菌的全过程.
