响应面法优化香猪骨制备BMP的脱钙工艺

摘要 为优化制备骨形态发生蛋白(BMP)过程中脱钙的工艺条件,以香猪骨为原材料,HCl溶液为脱钙剂,在单因素试验基础上,选择脱钙时间、HCl浓度和液料比(mL:g)为自变量,脱钙液中钙含量为响应值,利用响应面分析法,研究各自变量交互作用及对脱钙效果的影响。结果表明,香猪骨粉脱钙的最佳工艺条件为脱钙时间50 h,HCl浓度0.62 mol/L,液料比21.25(85 mL∶4 g)。在此条件下,脱钙液中钙含量为49.60 mg/100 mL。 In order to optimize the decaIcihcatlon technological conditions in the process oi BMk' preparation, taking the pig bone and the HCI solution as the raw material and decalcifying agent respectively, the effect of independent variables interaction on decalcification was studied by response surfaee methodology based on single factor experiment taking decalcification time, HC1 concentration, solid-liquid ratio as independent variables and calcium content in decalcification solution as the response value. The results showed that the optimum decalcification technological conditions included decalcification time 49.87 h, HC1 concentration 0.65 mol/L and solid-liquid ratio 21.25(85 mL : 4 g) respectively. The calcium content in decalcification solution was 49.60mg/100 mL under the optimum conditions.
