摘要
为小蓬竹增殖培养和生根培养提供参考,促进小蓬竹的保护及开发利用,用小蓬竹当年生半木质化枝条为外植体,进行了小蓬竹组织培养中消毒方法及诱导培养的研究。结果表明：将外植体在75%酒精中浸泡1 min,0.1%升汞溶液消毒10 min,无菌水冲洗5~6次消毒效果最好,成活率较高;采用MS培养基的诱导率达到73%,显著高于1/2 MS和1/4 MS培养基的诱导率;在不同激素组合下,6-BA浓度达到3.0 mg/L时诱导效果最好,当6-BA浓度高于3.0 mg/L时,其诱导率呈下降趋势。
By using semi-lignified branches of Drepanostachyum luodianense as explants,the disinfection method and inducing culture were studied to provide reference for multiplication culture and rooting culture of Drepanostachyum luodianense.The results showed that explants immersed in 75% alcohol for 1 minute,and then disinfected by 0.1% mercuric chloride for 10 minute,finally rinsed with sterile water for 5~6 times could reach good effect of disinfection and high survival rate.The induced effect of MS culture medium was significantly higher than that of 1/2MS culture medium and 1/4MS culture medium and the induction rate of MS culture medium reached 73%.With different hormone combination,induction rate increased with the increase of 6-BA concentration.When 6-BA concentration was 3.0mg/L,the induced effect was the best,but inductivity decreased when 6-BA concentration was higher than 3.0mg/L.
