摘要
以中国李品种"海湾红宝石"茎段为外植体,研究了不同光质对其茎段组织培养的影响。结果表明:在5种单一光照条件下,外植体的增殖倍数表现出极显著差异,在白光(60 d)条件下,外植体增殖倍数最高,为4.34倍;红光次之;蓝光和黑暗条件下较低。不同光质与白光组合或白光与不同光质组合对外植体增殖倍数有极显著影响,红光(30 d)—白光(30d)组合或者白光(30 d)—红光(30 d)组合能提高外植体增殖倍数,生长较快;其他光质组合条件下,外植体增殖倍数较低,长势差,嫩茎伸长缓慢,黄化现象严重,外植体逐渐死亡。说明"海湾红宝石"初代培养可选用单一的白光照射。
The stem segments of chinese plum(Prunus salicina Lindl.cv.Gulf-ruby) were used to study the effect of different light qualities on the tissue cultrue.The rusults indicated that the multiplication times was the highest(4.34) on single white light(60 d),effect of red light was next,and effects of blue light and darkness were worst.The Red(30 d)-White(30 d) combination or White(30 d)-Red(30 d) combination could increase the explant multiplication times,and the treated explants grew better.However,other light combinations induced lower multiplication times and worse growth,and the treated explants presented slow growth,etiolation and even death.It showed that Gulf-Ruby 1st generation culture might choose only white light.
