气虚血瘀状态下BALB/c小鼠结肠癌血行转移的机制研究

Study of the Influence of the QiXu and XueYu Syndrome on Artificial Hematogenous Metastasis of Conlon Cancer in BALB/c Rats

目的:研究气虚血瘀证对BALB/c小鼠结肠癌人工血行转移的影响及相关机制。方法:将BALB/c小鼠随机分为4组,即空白对照组、血瘀组、肿瘤组和瘀加瘤组。于利血平血瘀造模的第15天经尾静脉接种肿瘤,第28天处死小鼠检测全血黏度、细胞外信号调节激酶1/2(ERK1/2)、基质金属蛋白酶2(MMP-2)及基质金属蛋白酶12(MMP-12)。结果:血瘀组及瘀加瘤组全血黏度均明显升高,与空白对照组比较差异极显著(P<0.01)。瘀加瘤组右肺转移灶计数明显高于肿瘤组,差异有显著性(P<0.05)。ERK1/2、MMP-2按血瘀组、肿瘤组、瘀加瘤组顺序梯度递增,与空白对照组比较差异显著(P<0.05)。MMP-12的表达在血瘀组减弱,瘀加瘤组增强,组间差异有显著性(P<0.05)。结论:气虚血瘀证能促进结肠癌转移,其作用机制可能是通过上调ERK1/2和MMP-2的表达,同时抑制MMP-12的表达来完成。

Objective：To study the mechanism about artificial hematogenous metastasis of colon cancer which affected by QiXu and XueYu syndrome.Methods：Balb /c rats were randomly divided into four groups：control group,stasis group,tumor group,stasis and tumor group.Detect the whole XueYu,ERK1/2,MMP-2 and MMP-12 on the 28th day.Results：The whole XueYugroup,XueYu and tumor group were more significantly improved than control group（P0.01）.The amounts of metastasis tubercle on lung in XueYu and tumor group were more significantly improved than tumor group（P0.05）.The protein expression of ERK1/2 and MMP-2 increased in order of XueYu group,tumor group,XueYuand tumor group which were significantly strengthened than control group（P0.05）.The proteolytic activity of MMP-12wasmuch weakened in XueYu group,and much strengthened in XueYu and tumor group（P0.05）.Conclusion：The influence of QiXu and blood XueYu syndrome could promote the hematogenous meta-stasis of colon cancer.The mechanism was probably related to the enhancement of the expression of ERK1/2 and MMP-2,and restrain the expression of MMP-12.
