正电子显像剂^(18)F纳米羟基磷灰石制备

摘要 目的制备18F标记的纳米羟基磷灰石,以期开发一种PET-CT显影剂,为今后研究羟基磷灰石体内分布、抑制肿瘤的机理提供基础材料。方法采用溶胶-凝胶法合成纳米羟基磷灰石,在合成过程中加入正电子核素18F,通过置换羟基磷灰石中的部分OH-,获得标记有18F的羟基磷灰石显影剂。通过薄层扫描测得标记物的放化纯度,并进行了红外光谱和X射线衍射测定。结果合成的纳米羟基磷灰石的放化纯度均在95%以上,符合实验要求,且具有较好的体外稳定性。红外光谱和X射线衍射测定表明18F已取代部分OH-。结论制备成功18F标记的纳米羟基磷灰石。 Objective To prepare 18F-labeled hydroxyapatite nanoparticle in order to develop a PET-CT imaging agent for further research on the distribution of hydroxyapatite in vivo and its possible mechanisms of tumor suppression.Methods Nano-hydroxyapatite was synthesized using sol-gel method.During this period,18F-labeled hydroxyapatite imaging agent was obtained by adding positron 18F to replace partial hydroxyl in nano-hydroxyapatite.Its radiochemical purity was measured by thin-layer chromatography(TLC).Infrared spectrum and diffraction of X-ray(XRD) patterns were tested as well.Results The radiochemical purity of synthesized nano-hydroxyapatite with good stability in vitro was above 95%,which met the experimental requirement.Infrared spectrum and XRD patterns showed that partial hydroxyl had been alternatived by 18F.Conclusion 18F-labeled nano-hydroxyapatite is successfully synthesized.It can provide a primary material for the study of hydroxyapatite.
