持续性心房颤动患者心房肌中丝/苏氨酸蛋白磷酸酶活性变化及其意义

摘要 目的检测持续性心房颤动(简称房颤)患者心房肌中丝/苏氨酸蛋白磷酸酶的活性变化并探讨其意义。方法用荧光法检测16例持续性房颤及14例窦性心律患者右心耳心肌组织中三种丝/苏氨酸蛋白磷酸酶的活性,同时采用Western blot技术检测第16位点丝氨酸磷酸化的受磷蛋白表达情况。结果与窦性心律组相比,持续性房颤组1型、2A型和2B型丝/苏氨酸蛋白磷酸酶活性均明显增加(P<0.01),第16位点丝氨酸磷酸化的受磷蛋白表达水平明显降低(P<0.01),且与1型和2A型丝/苏氨酸蛋白磷酸酶活性成负相关。结论持续性房颤患者心房肌丝/苏氨酸蛋白磷酸酶活性明显增加,其可能通过影响受磷蛋白磷酸化水平在心房电重构过程中发挥作用。 Objective To investigate the possible mechanism of electrical remodeling in persistent atrial fibrillation (PAF) by evaluating the changes of the Serine/Threonine protein phosphatase ( Ser/Thr PP) activities in atrial tissue in patients with PAF. Methods The Ser/Thr PP activities in fight atrial appendage tissue obtained from 16 patients with PAF and 14 patients with sinus rhythm(SR) were tested by use of ProFluor Ser/Thr PPase Assay while the phosphorylation level of 16-point serine residues of phospholamban(PLBp-Ser16) were tested by use of Western blot analysis. Results The Ser/Thr PPs activities were higher in patients with PAF than those in patients with SR (P 〈 0.01 ) ; The PLBp-Sert6 in patients with PAF were lower than that in patients with SR ; Both type 1 Ser/Thr PP ( PP1 ) activity and type 2A Ser/Thr PP ( PF2A ) activity correlated negatively with the level of PLBp-Ser16 in patients with PAF( P 〈 0. 01 ). Conclusions The Ser/Thr PPs activities increase in patients with PAF , which may play a role in electrical remodeling in PAF through influencing phosphorylation of phospholamban.
