摘要 研究红曲代谢产物Monascin对人恶性黑色素瘤细胞A375的诱导分化作用。用MTT法评价Monascin对A375增殖的抑制作用;PI流式单染检测细胞周期分布;透射电镜观察细胞超微结构变化;以黑色素含量、酪氨酸酶活力和体外迁移能力为指标检验Monascin对A375诱导分化作用。Monascin对A375具有一定的增殖抑制作用,并能将其阻滞在G0/G1期;透射电镜观察到A375核质比变小且细胞器增多;黑色素含量增多,相对酪氨酸酶活力提高,体外迁移能力下降。试验结果表明,Monascin对A375具有较显著的诱导分化作用。 To study the differentiation effect of Monascin on human malignant melanoma cell A375,the inhibitory effect of cell proliferation was tested by MTT assays.The cell cycle distribution was detected by Flow Cytometery.The morphology of A375 cells treated with Monascin was detected by TEM.The melanin content and total activity of tyrosinase of A375 cells were assessed after the treatment with Monascin.The cell migration was determined by transwell chamber model.The result showed that A375 can be inhibited by Monascin,showing a time-and dose-dependent manner,the cell cycle of the A375 cells treated with Monascin was arrested in G0/G1 phase.The ratio of nucleus/cytoplasm was decreased and the enrichment of cytoplasmic organelle was also observed by TEM.Moreover,the Monascin could promote A375 cells' melanin biosynthesis,and up-regulate its total tyrosinase activity but decreased its migrant ability in vitro.The Monascin can induce differentiation of A375,although the mechanisms need to more the further research.
