口腔鳞癌组织CK17蛋白与mRNA水平的表达及其临床意义

摘要 目的:探讨口腔鳞癌组织标本中细胞角蛋白17(cytokeratin 17,CK17)的蛋白与基因水平的表达及其临床意义。方法:在30例口腔鳞癌患者的癌组织及癌旁组织标本中,分别采用免疫组织化学和荧光实时定量RT-PCR方法,检测CK17的蛋白表达和mRNA水平,并与各临床病理指标进行统计学分析,采用SPSS10.0软件包对数据结果进行统计分析。结果:口腔鳞癌组织标本中CK17蛋白和mRNA表达水平均显著高于癌旁组织(P<0.01)。CK17的表达水平与烟酒嗜好、肿瘤大小、淋巴结转移、临床分期、病理分化程度无显著相关性。结论:口腔鳞癌组织中CK17蛋白质和mRNA表达上升,提示其与肿瘤的发生、发展具有一定关系。 Objective: To detect the protein expression and gene transcript level of cytokeratin 17 (CKl7) in human oral squamous cell carcinoma tissues and explore their clinical significance. Methods: The protein expression and mRNA level of CK17 in 30 cases of oral squamous cell carcinoma tissues and paracancerous tissues were detected by immunohistoehemistry and fluorescent quantitive real-time RT-PCR techniques respectively. The relationship between the CK17 expression level and the clinicopathologieal parameters was analyzed by SPSS10.0 software package for analysis. Results: The CK17 protein expression and mRNA transcript level in cancerous tissues increased significantly comparing to the para-cancerous tissues. No significant correlation was found between the CK17 expression and clinical status, such as smoking, drinking, T stage, N stage, clinical stage and pathological differential degree. Conclusion: These results suggest that increased CK17 expression in OSCC tissue specimens could contribute to carcinogenesis process of OSCC, which provides some basis for further study on the carcinogene sis mechanism of OSCC.
