摘要 目的探讨位于脂氧酶12(LOX12)基因启动子区的-183G>A单核苷酸多态与胃癌发病风险之间的关系。方法以聚合酶链反应-限制性片段长度多态分析方法,检测100例胃癌患者和119例无肿瘤对照组的基因型。以Logistic多因素回归模型计算各基因型的胃癌风险。结果 LOX12-183G和-183A等位基因频率在对照组及胃癌组中分别为59.7%、40.3%和48.0%、52.0%,两组比较差异有统计学意义(P<0.05)。与GG基因型携带者相比,AA基因型显著增加胃癌发病风险[OR=2.36;95%CI(1.12,5.70)]。结论 LOX12启动子区-183G>A变异是胃癌的重要遗传易感因素,在胃癌的发生发展中发挥着重要的作用。 Objective To explore the relationships between 12-lipoxygenase(LOX12) promoter-183G/A polymorphism and the risk of developing gastric cancer(GC).Methods One hundred cases of gastric cancer and 119 cancer-free controls were genotyped for the LOX12-183 polymorphism by PCR-RFLP method.The odds ratios(ORs) and 95% confidence intervals(CIs) were calculated using unconditional Logistic regression model.All statistical tests were two-sided.Results The frequency of-183G allele and-183A allele were(59.7% vs.40.3%) and(48.0% vs.52.0%) in controls and in GC patients,respectiveily(P0.05).The-183 AA genotype was related to an increased risk for GC[OR=2.36,95%CI(1.12,5.70)],compared with the GG genotype.The impact of-183G/A polymorphism on GC pathogenesis existed in a dose-response manner.Conclusion The LOX-183G/A SNP polymorphism might be an important genetic predisposing factors for the development of gastric cancer.
