摘要
目的检测Paxillin在胃腺癌(GAC)、癌前期变化及慢性浅表性胃炎(CSG)中的表达,探讨其生物学功能及其与GAC病理特征的关系。方法免疫组织化学链霉菌抗生物素蛋白-过氧化物酶(S-P)法检测59例GAC、30例CSG及80例癌前期变化[包括30例慢性萎缩性胃炎(CAG)、30例肠上皮化生(IM)和20例异型增生(Dys)]组织中Paxillin的表达。结果 Paxillin在CAG组、IM组、Dys组和GAC组中表达均高于CSG组,差异有统计学意义(P<0.05)。Paxillin的表达程度与胃黏膜病变程度呈正相关(rs=0.779,P=0.000)。Paxillin在进展期、伴淋巴结转移及中、低分化组GAC组织表达高于早期、无淋巴结转移及高分化组GAC组织,差异均有统计学意义(P<0.05)。结论 Paxillin表达增高是GAC癌变和转移的重要分子事件,其影响可能在GAC发生的早期即已存在,并持续到GAC阶段,有望成为GAC淋巴结转移的预测指标。
Objective To investigate the expression levels of paxillin in gastric adenocarcinoma(GAC),preneoplastic changes and chronic superficial gastritis(CSG) tissues,and to study its possible roles in the carcinogenesis and metastasis of GAC.Methods The expression levels of paxillin in tissues of GAC(59 cases),CSG(30 cases) and preneoplastic lesions(80 cases) were determined using S-P immunohistochemical techniques.The preneoplastic lesion group included chronic atrophic gastritis(CAG,30 cases),intestinal metaplasia(IM,30 cases),and dysplasia(Dys,20 cases).Results The expression levels of paxillin in CAG,IM,Dys and GAC tissues were significantly higher than those in CSG tissues,with the highest expression found in GAC tissues(P0.05).The expression levels of paxillin were positively correlated to severity of the gastric mucosal lesions(rs=0.779,P=0.000).The expression levels of paxillin in tissues from patients with advanced GAG,lymph node metastasis,and moderately and poorly differentiated GAC were significantly higher than in those from patients in an early GAG stage,with no lymph node metastasis,and with well-differentiated GAC(P0.05).Conclusion The up-regulation of paxillin expression is an important molecule event in the carcinogenesis and metastasis of gastric adenocarcinoma,the influence of which may emerge in the early stage of carcinogenesis and continue to the advanced stage.Our findings support the use of paxillin as a molecular index for prediction of lymph node metastasis.
