环孢A处理对大鼠横断脊髓cdk4表达的影响

摘要 目的探讨环孢霉素A处理对大鼠横断脊髓cdk4表达的影响.方法成年SD大鼠分SCT组(T9横断),和SCT后给予环孢霉素A处理组.每组13只动物.动物于环孢霉素A处理7 d后处死,其中每组5只用免疫组化染色确定cdk4在脊髓的定位分布,另8只动物用RT-PCR技术确定cdk4在损伤脊髓的表达变化.结果与手术组比较(0.99±0.11),环孢霉素A处理(1.12±0.15)导致cdk4在损伤脊髓的表达明显上调(P<0.05);免疫组化染色显示cdk4主要分布在脊髓灰质神经元.结论环孢霉素A处理能有效上调损伤脊髓cdk4表达水平,提示环孢霉素A发挥免疫抑制作用可能与cdk4的调节有关. Objective To explore the effects of CSA on the cdk4 expression in the transected spinal cord of rats.Methods Sprague-Dawley(SD)rats were randomly divided into operation group(T9 transection)and CSA treatment group 13 rats in each group.After spinal cord transection(SCT)was performed,CSA was intrapentoneously injected one times every day till sacrificed animal on 7 days following SCT.After spinal cords(n=8)were harvested,RT-PCR was used to detect the level of cdk4(β-actin was used as control) in 8 rats.The left 5 animals in each group were subjected to immunohistochemical staining to determine the localization of cdk4.Results Following CSA treatment,the expressional levels of cdk4 at 7day post operation increased more significantly than in SCT rats(P〈0.05).The immunoreactive stain of cdk4 was mainly seen in the cytoplasm of neurons in the gray matter.Conclusion CSA can upregulate the expression of cdk4 that located in neurons in injured spinal cord,indicating that CSA may play a role in neuroprotective effect that could be associated with cdk4 regulation in rats subjected SCT.
