环孢A处理对脊髓损伤大鼠后肢骨骼肌RAF-1表达的影响

Effects of Cyclosporine A on RAF-1 Expression in Skeletal Muscles of Hindlimbs in Adult Rats after Spinal Cord Transaction

摘要 目的探讨环孢霉素A处理对脊髓损伤大鼠后肢骨骼肌RAF-1 mRNA表达的影响.方法成年SD大鼠分SCT组(T9横断),和SCT后给予环孢霉素A处理组.脊髓横断组用环孢霉素A处理(1 mg/200 g)7 d后处死动物.取后肢骨骼肌用免疫组化染色(n=5)确定RAF-1 mRNA在后肢骨骼肌的定位分布.RT-PCR技术检测RAF-1 mRNA在损伤脊髓大鼠后肢骨骼肌的表达变化(n=8).结果与手术组比较(0.95±0.13),环孢霉素A处理(0.75±0.14)导致RAF-1 mRNA在损伤脊髓大鼠后肢骨骼肌的表达明显下调(P<0.05);RAF-1蛋白产物主要分布在细胞浆.结论环孢霉素A处理能有效下调脊髓损伤大鼠后肢骨骼肌RAF-1 mRNA表达水平. Objective To investigate the effects of cyclosporine A on RAF-1 expression in the skeletal muscles of hindlimbs in adult rats after spinal cord transaction.Methods Sprague-Dawley(SD)rats were randomly divided into operation group(T9 transection)and CSA treatment group.After spinal cord transection(SCT)was performed,CSA(1 mg/200 g)was intrapentoneously injected one time every day to rats in CSA treatment group till 7 days after SCT.The spinal cords were harvested,then RT-PCR(n=8)and immunohistochemistry(n=5)were respectively performed to detect the levels of RAF-1 mRNA and localization of raf protein in the skeleton muscle.Results After CSA treatment,a significant decrease in the expression levels of RAF-1 mRNA(0.75 ±0.14)was found at 7day post operation,compared with that of the SCT rats(0.95±0.13)(P〈0.05).RAF-1 protein was distributed in the cytoplasma.Conclusion CSA can downregulate the expression of RAF-1 mRNA in skeleton muscles of hindlimbs in rats following spinal cord injury.
