摘要
应用PCR 技术从鸡肝脏基因组中克隆了1.1 kb 的鸡卵清蛋白基因上游调控序列,并进行了序列分析。结果发现,扩增产物只有2 个碱基发生了突变,其TATA 框、组织特异性因子和卵清蛋白上游启动子均未发生变异,表明已成功地克隆了鸡卵清蛋白基因上游调控序列,这为应用卵清蛋白提取生物活性物质的转基因鸡的构建研究奠定了基础。
The upstream regulatory regions of 1.1 kb(-1 047^+39) of chicken ovalbumine gene were amplified by PCR from chicken liver DNA and sequenced .It was showed that two bases mutations occurred in some sites by comparison with the published sequence. The regulatory sequences as a promoter might be used to target some valuable foreign gene into and expressed in the chicken ovalbumine.
