摘要 目的探讨银杏叶提取物(Egb761)对哮喘大鼠气道上皮细胞基质金属蛋白酶-9(mmp-9)表达的影响及对气道炎症的干预作用。方法将SD大鼠随机分为4组,即正常对照组(A组),哮喘模型组(B组),地塞米松组(C组),Egb761组(D组),建立大鼠哮喘模型,用免疫组化技术结合计算机图像分析系统,测定各组大鼠气道上皮细胞mmp-9表达的相对含量,同时检测支气管肺泡灌洗液(BALF)中嗜酸性粒细胞(EOS)计数及细胞分类计数。结果①mmp-9表达的相对含量,哮喘模型组明显高于正常对照组(P<0.05);Egb761组明显低于哮喘模型组(P<0.05)且与地塞米松组差异无统计学意义(P>0.05);②Egb761组BALF中EOS的表达明显低于哮喘模型组(P<0.05)且与地塞米松组差异无统计学意义(P>0.05)。结论 Egb761可以减少哮喘大鼠气道上皮细胞mmp-9的表达,同时对气道的炎症有明显的干预作用。 Objective To investigate the effects of ginkgo bioba extract(Egb761) on matrix metalloproteinase-9(mmp-9) expression and evaluate the intervention effect on airway inflammation in asthmatic rats model.Methods 40 SD rats were randomly divided into 4 groups: normal control group(group A),asthmatic model group(group B),dexamethasone group(group C) and Egb761 group(group D).The models of asthma were established by repeated inhalation of ovalbulium.The expression of MMP-9 in bronchial epithelium in these groups were determined using immunohistochemical technique and computer assisted image analysis system.The Eos amounts and differential leukocytes counts in bronchoalveolar lavage fluid(BALF) were counted.Results ①The expression of mmp-9 in the epithelial cells of bronchi was significantly higher in the asthmatic group than normal control group(P0.05).The expression of mmp-9 in Egb761 group was much lower than that in asthmatic model group(P0.05).There were no statistical significant difference between dexamethasone group and Egb761 group(P0.05).②The Eos amounts in BALF of Egb761 group was much lower than that in asthmatic model group(P0.05).There were no statistical significant difference between dexamethasone group and Egb761 group(P0.05).Conclusion Egb761 could inhibit the expression of mmp-9 and decrease airway inflammation in asthmatic rats.
