白细胞介素-10对风湿性心脏病患者外周血单个核细胞NF-κB/p65表达的影响

Effects of Interleukin-10 on expression of nuclear factor kB/p65 on peripheral blood mononuclear cell in rheumatic heart disease patients

目的探讨白细胞介素-10(IL-10)对风湿性心脏病患者(RHD)外周血单个核细胞(PBMCs)核转录因子appaB(NF-κB)/p65表达的干预作用及NF-κB与心功能的关系。方法 22例RHD患者和15名健康对照者进行心脏超声心动图检查,并对所有入选者分离出的PBMCs进行培养,并将培养的PBMCs分成3组:空白对照组、脂多糖(LPS)组和白细胞介素-10组。对3组培养后的PBMCs NF-κB/p65的表达进行免疫组化染色检测和半定量分析。结果 RHD患者PBMCs NF-κB/p65胞核染色阳性率明显高于健康对照者[(24.8±8.6)%比(8.2±3.4)%,P<0.01];RHD患者随着心功能恶化NF-кB/p65表达水平逐渐升高;脂多糖组RHD患者和健康对照者PBMCs NF-κB/p65胞核染色阳性率与空白对照组比较均显著增加(P<0.01);白细胞介素-10组RHD患者和健康对照者PBMCs NF-κB/p65胞核染色阳性率与脂多糖组比较则显著下降(P<0.01)。结论 RHD患者NF-κB/p65表达增高,而白细胞介素-10可下调脂多糖刺激下NF-κB/p65的表达增高;它的检测有助于对RHD患者心功能恶化的判断。

Objective To investigate the expression of nuclear factor kB/p65 on peripheral blood mononuclear cells（PBMCs）in Rheumatic heart disease（RHD） patients and effect of Interleukin-10（IL-10） and the relation with cardiac function.Methods All patients undertook ultrasonic cardiography.PBMCs isolated from peripheral blood in RHD patients（n=22）and healthy subject（n=15）were divided into 3 groups：control group,lipopolysaccharide（LPS） stimulate group and IL-10 group,The expression of NF-κB/P65in PBMCs were measured by immunocytochemical staining.Results The expression of NF-κB/P65 is increasing with heart function failure in RHD patients.Compared with the healthy subjects,the expression of NF-κB/P65 on PBMCs in RHD patients was found significantly increased [（24.8±8.6）%比（8.2±3.4）%,P0.01].It is increasing with heart function failure in RHD patients.In LPS stimulation,the expression of NF-κB/P65 on PBMCs in RHD patients and healthy subject was found significontly increased（P0.01）.IL-10 significantly inhibited this increment in RHD patients and healthy subject（P0.01）.Conclusion The expression of NF-kB/P65 in RHD patients is found significantly increased,but IL-10 can inhibit LPS stimulating increment of NF-κB/P65.It may be a predictor of the severity of heart remodeling and heart function failure.
