摘要 目的:建立高效液相色谱法测定黄瓜皮中绿原酸含量的方法。方法:采用Zorbax Eclipse XDB-C18色谱柱(150×4.6 nm,5μm);流动相:甲醇-0.4%磷酸溶液(25∶75);流速:1.0 ml.min-1;检测波长:326nm;柱温:30℃。结果:绿原酸的线性范围为0.0111~0.1332 g.L-1(r=0.9995),平均加样回收率(n=5)为99.97%,RSD为2.07%,结果显示黄瓜皮中绿原酸的质量分数均大于8.10 mg.g-1。结论:本方法简便快捷、精密度高、重现性好,可用于黄瓜皮中绿原酸含量的测定。 To establish an HPLC method for determination of chlorogenic acid in cucumber skin,Zorbax Eclipse XDB-C18 was adopted.The mobile phases consisted of CH3OH-0.04% H3PO4(25∶75) at a flow rate of 1.0 mL·min-1,the detection wavelength was set at 326nm,and column temperature was 30 ℃.The linear range of chlorogenic acid was 0.0111~0.1332mg·mL-1(r= 0.9992).The average recoveries(n=5) was 99.97% and RSD was 2.07%.The contents of chlorogenic acid in cucumber skin were more than 8.10 mg·g-1.The method was simple,rapid and accurate with a good reproducibility,and could be applied to determine of chlorogenic acid in cucumber skin.
