摘要
探讨白细胞介素-11(IL-11)及乙醇对人肝肿瘤细胞系(HepG2)载脂蛋白B(apoB)mRNA表达及蛋白分泌的作用。培养HepG2细胞,分组加入不同浓度IL-11进行浓度试验,收集培养液及细胞。用火箭电泳法及Northernblot分别检测培养液中apoB含量及HepG2细胞apoBmRNA的表达。IL-11及乙醇抑制试验按同法分组进行。结果IL-11能抑制HepG2细胞apoB表达与分泌,且呈量效正相关。IL-11能明显抑制乙醇的正向作用。说明IL-11能调节HepG2细胞apoBmRNA表达与分泌,可参与调节机体脂质代谢。
To study the effects of interleukin (IL)-11 and ethanol on the expression and secretion of apoB in HepG2 cells. HepG2 cells were cultured in DMEM and divided into 4 groups with addition of different concentrations of IL-11. Medium was collected for measuring apoB by using electroimmunoassays, and cells harvested to detect the expression of apoB mRNA by using Northern blot analysis. Ethanol inhibiting experiment by IL-11 was performed with the same methods. It was found that IL-11 could inhibit the expression of apoB mRNA and its secretion in HepG2 cells in a dose-dependent manner and obviously inhibit the positive action of ethanol, suggesting that IL-11 could regulate the expression and secretion of apoB in HepG2 and may play some role in the apolipoprotein metabolism of liver cells.
