摘要
根据Rosenberg 法复制脑出血大鼠模型;采用细胞形态学、DNA 裂解率、流式细胞仪方法,观察脑出血损伤后,大鼠海马CA1 区神经元的形态结构,DNA 完整性,神经细胞发生凋亡的数量;结果,实施造模手术的大鼠清醒后,均出现偏瘫等神经系统病态体征。造模7 天时,脑溢安颗粒剂组(以下简称NYA组)CA1 区锥体细胞形态基本正常,模型组CA1 区仍可见到多量的凋亡锥体细胞,差异具有显著性意义(P< 0.01);DNA裂解率测定,在1 d、4 d、7 d 3 个时间点上NYA 组DNA 裂解率比模型组低10% ~14% ;流式细胞仪检测,造模后7 d,NYA组神经细胞凋亡低于模型组,差异具有显著性意义(P< 0.05)。本研究提示,NYA具有抑制神经细胞DNA断裂,减少神经细胞凋亡的作用。
Cerebral hemorrhagic rat models were made according to Rosenberg method and the morphosis of neuron in CA 1 region,DNA integrity and the number of withered nerve cells were observed.The results showed that every rat appeared hemiplegic;7 days later,the morphosis of vertebral cell in region CA 1 was almost normal in Naoyi'an group and mostly withered in model one with signifcant differency(P<0.01);DNA cleavage rate in Naoyi'an group was 10%-14% lower than that of model group;the withering of nerve cell in Naoyi'an group was lower than that of model group with signifcant differency(P<0.05).It suggested that Naoyi'an granule could restrain the breaking of DNA and the withering of nerve cell.
