摘要
目的临床验证银杏叶提取物(ginkgo biolba extract,EGB)对急性脑梗死患者血小板磷酸二酯酶3(phos-phodiesterase3,PDE3)活性及其相关因素的影响,观察其临床疗效。方法 30例急性脑梗死患者为对照组,接受常规治疗。15例急性脑梗死患者,常规治疗外给予EGB治疗。分别于用药前、用药后第1、3、7、14天测定两组患者血小板PDE3活性、环腺苷酸(cylic adenosine monophosphate,cAMP)水平、胞浆游离钙离子([Ca2+]i)水平,并于用药前及用药后第14天进行神经功能缺损评分、血液流变学检查,作为临床疗效评价指标。结果 EGB可使急性脑梗死患者血小板PDE3活性进一步下降,大大提升cAMP水平,降低胞浆[Ca2+]i浓度,改善血液流变学及神经功能缺损情况。结论 EGB可使急性脑梗死患者血小板PDE3活性进一步下降,加强PDE3在脑梗死急性期的保护作用,有效抑制血小板聚集,改善血液循环及神经功能缺损。
Objective To observe the effect of ginkgo biolba extract(EGB) on phosphodiesterase 3(PDE 3) activity and the correlative factors in patients with acute cerebral infarction,and verify the therapeutic effect of EGB.Methods Fifteen patients with acute cerebral infarction received EGB administration besides common therapy.Control group(30 patients with acute cerebral infarction) received common therapy.Platelet PDE 3 activity,cylic adenosine monophosphate(cAMP) levels and i concentration of both groups were detected in various timepoints(before treatment and the 1st,3rd,7th,14th day after treatment).Clinic indexes(neurologic impairment and hemorheology) were evaluated before treatment and at 14th day after treatment.Results EGB inhibited the platelet PDE 3 activity of patients with acute cerebral infarction,greatly increased the cAMP levels,lowered the i concentration,and improved the hemorheology and neurologic impairment of the patients.Conclusion In acute stage of cerebral infarction,EGB can significantly inhibit patients' platelet PDE 3 activity,increase cAMP,decrease i,and relieve clinical symptoms.
