摘要
大量研究表明,长期饮茶尤其绿茶者,可大大降低各种癌变的发病率,而且对多种体内外肿瘤具有直接杀伤作用.作者通过采用不同浓度的茶多酚作用于体外培养的肝癌细胞株Hep-G2细胞,初步探讨茶多酚对Hep-G2细胞的凋亡作用.采用MTT法(噻唑蓝比色法)和AO-PI(吖啶橙-碘化丙啶)双重荧光染色法研究了茶多酚诱导Hep-G2细胞株的凋亡作用.MTT法研究结果表明,在一定的浓度范围内,茶多酚对Hep-G2细胞有明显的抑制作用,且存在良好的剂量效应关系,随着茶多酚浓度的升高,其对Hep-G2细胞的抑制率也升高.Hep-G2细胞经茶多酚作用后,细胞形态发生明显的变化,可见凋亡小体,荧光强度增强,细胞呈现凋亡现象.茶多酚在体外对Hep-G2细胞的增殖具有抑制作用,诱导其发生凋亡.研究结果为茶多酚的进一步开发应用奠定理论基础.
Many studies indicated that long-term tea drinking,especially the drinking of green tea,could not only reduce the incidence of various cancers greatly,but also have direct killing effects on a variety of tumor in vivo and in vitro.To verify the effects of tea polyphenols(TP) on apoptosis in vitro,the effects of TP on apoptosis of Hep-G2 cell were evaluated by MTT(MTT assay) and AO-PI(acridine orange-propidium iodide) double fluorescence staining.The results of MTT assay showed that in a certain concentration range,TP inhibited significantly the growth of Hep-G2 cell,and the inhibitory effect showed good dose-response relationship with the TP concentration.With raising the TP concentration,its inhibitory rate of Hep-G2 cell increased.After treated by TP,morphological changes of Hep-G2 could be observed obviously,with apoptotic bodies appeared and fluorescence intensity enhanced,which indicated that apoptosis occurred in these cells.The results suggested that TP could inhibit the proliferation of Hep-G2 and induce its apoptosis in vitro.This work will give rise to the application of TP in cancer prevention.
