摘要
目的 观察前胡丙素(praeruptorin C,Pra- C)对肾血管性高血压(renalhypertension.RH) 模型大鼠主动脉平滑肌中AT1受体变化的影响。方法 两肾一夹Wistar 大鼠,术后5 周给予Pra - C〔20 mg/(kg·d) ,ig〕,观察大鼠主动脉的血管紧张素(AngⅡ)受体第一亚型(AT1) 的反应性。结果 Pra- C明显降低RH 大鼠血压和主动脉对Ang Ⅱ的反应性,用药2 周、4 周后血压明显下级,分别由治疗前的(23-6 ±2-1) 降到(18-6 ±1-8)和(15-8±1-9) kPa(P< 0-01) ;主动脉对Ang Ⅱ的最大收缩反应(Emax) 较对照组(863-16 ±41-07)mg 明显为低,分别为(547-27±34-50)mg,(483-16 ±31-26)mg(P< 0-01) ,量效曲线向右下移位。结论 Pra- C 降低血压作用与主动脉AT1 受体反应性下降有关。
Objective To observe the effects of praeruptorin C on blood pressure and responsiveness of angiotensin Ⅱ-1 receptor of aortic artery in renal hypertension rats. Method Two-kidney one-clip Wistar hypertensive rats were treated with praeruptorin C 〔Pra-C,20 mg/(kg·d),ig〕for 2 and 4 weeks and the response of aortic artery to angiotensin were studied. Results Pra-C significantly decreased the blood pressure and the response of aortic artery to angiotensin Ⅱ. The blood pressure at 2 and 4 weeks after administration decreased from the control of (23.6±2.1)kPa to (18.6±1.8) kPa and (15.8±1,9)kPa ( P<0 01 );the Emax of vasoconstriction to angiotensin Ⅱ decreased from (863.16±41.07) mg to (547.27±34.50)mg and (483.16±31.26) mg ( P <0 01),with doseresponse curves shifting rightward. Conclusion The effects of Pra-C on blood pressure are related to its effects on the responsiveness of aortic artey AT 1 receptors in renal hypertensive rats.
