不同被毛密度獭兔的皮肤组织差异表达基因研究 被引量:2

摘要 为了筛选出不同被毛密度獭兔皮肤组织中差异表达的基因,本研究采用Agilent公司家兔全基因表达谱芯片对不同被毛密度獭兔皮肤组织总RNA进行了芯片杂交,通过实时定量PCR对部分差异表达基因进行了验证,并对基因表达谱进行了分析。结果表明,芯片试验共检测到2 657个差异表达的基因,其中包含1 106个已知功能的基因,表达上调的基因687个,表达下调的基因419个。GO分析表明,参与细胞增殖、分化、凋亡,信号传导及离子转运等生命过程的多种基因的表达水平发生了明显改变。实时定量PCR所得结果与基因芯片结果基本吻合。结果说明不同被毛密度獭兔皮肤组织的基因表达谱存在差别,这些基因的异常表达,可能参与皮肤毛囊的形成或分化过程,最终导致獭兔被毛密度的差异。这些基因的发现为进一步寻找与被毛密度相关的分子标记,并最终实现獭兔生产中的早期选育提供了研究方向。 In order to screen differentially expressed genes in Rex rabbit with different wool density,by using Agilent's Rabbit Gene Expression Microarray,the total RNA isolated from the rabbit's mid-dorsal skin samples was used to identify the genes differentially expressed in Rex rabbit with different wool density.The expression patterns of selected differentially expressed genes were further identified by quantitative real-time PCR.The results showed that 2 657 differentially expressed genes were identified.Among them,the function of 1 106 genes were known.The expression of 687 genes were up-regulated,and 419 genes were down-regulated.GO analysis results revealed that the expression levels of the most differentially expressed genes were related to cell proliferation,apoptosis,differentiation,signal transduction and ionic transport altered most.The alteration of gene expression from array data was confirmed by quantitative real-time PCR.The results suggest that the genes expression profile of Rex rabbit with different wool density is different,their inabnormal expression may involve in the formation and differentiation of skin follicle,and some important genes identified may be useful in further study on wool density molecular markers of Rex rabbit.
