摘要 研制了一种高灵敏度、高准确性、微小取样(一滴血,V≥ 25 μL)并可快速测量(≤60 s)的实用型夹心式胆固醇生物传感器。它是由双介体、三酶生化氧还体系构成的电流型传感器。其中,第一介体含于敏感电极石墨电导层内,第二介体及三种酶、相伴试剂含于试剂纤维层中。本传感器可用来直接测量从正常人到异常人的血胆固醇含量(2~10m m ol/L),并将实现商品化。 An amperometric, highly sensitive, accurate and fast response cholesterol biosensor was developed. This novel biosensor technology is based on a bi mediator, tri enzyme redox system. An innovative, truly “sandwich” sensor configuration significantly enhances its sensitivity. First redox mediator is directly present in the sensing electrode graphite layer. The second redox mediator and the enzymes (cholesterol esterase, cholesterol oxidase and horseradish peroxidase) with other necessary stabilizer and non reactive reagents are present in a highly water absorbing cellulose matrix. An applied voltage of -125mV provides added sensitivity and precision. The cholesterol biosensor only requires one drop of blood (≥25μl). Red blood cells are excluded by a blood separation membrane allowing only plasma to penetrate into the reaction center. Test results are displayed on an electronic meter after 60 s. This biosensor can directly measure cholesterol at the level range of 2~10 mmol/L (90~400mg/L),covering normal and abnormal ranges in humans. The handheld, battery operated compact unit is specifically designed for ease carry and commercialization and will benefit to the Coronary Risk Assessment Program for the general public.
