银杏叶提取物对大鼠脑出血后神经元保护作用的研究 被引量:15

摘要 目的:探讨银杏叶提取物对大鼠脑出血(ICH)周围神经元的保护作用。方法:胶原酶注入大鼠尾壳核建立ICH模型,然后将大鼠随机分为脑出血后治疗组,模型组和正常对照组。模型组和治疗组动物均于注射后6、24 h,3、7、14 d后处死。对各组大鼠进行神经功能评分;用干/湿重法测定脑组织的含水量;透射电镜观察神经元的超微结构并对线粒体进行体视学分析。结果:治疗组大鼠神经功能评分较模型组明显增加(P<0.05);模型组大鼠脑出血后24 h脑含水量开始升高,72 h达到高峰。银杏叶提取物治疗组脑含水量较模型组明显下降(P<0.05);与模型组相比治疗组神经元线粒体的数量明显增多(P<0.05);线粒体肿胀明显减轻(P<0.05)。结论:银杏叶提取物能保护神经元,促进神经功能恢复。 Objective: To study the protection of Leave Ginkgo Biloba extract on neurons that surround intracerebral hemorrhage(ICH)in rats.Methods: Collagenase was injected into caudate nucleus to induce rat model of ICH.The rats were randomly divided into 3 groups,treatment group after ICH,ICH modal group and normal control group.The rats of modal and treatment groups were sacrificed on 6,24 h and 3,7,14 days after injection.The score of nerve function to evaluate behavioral change;the dry/wet weight method to check up cerebral water content.The ultrastructure of neurons that surround ICH in rats was observed by transmission electron microscope and the mitochondria was performed a stereology analysis.Results: The score of nerve function was obviously increased after ICH in Leave Ginkgo Biloba extract treatment group,and had significant difference compared with ICH group(P0.05).The content of edema in rats in ICH group 24 h increased and were the highest at 72 h.The content of edema in the extract of Leave Ginkgo Biloba treatment group had significant decrease compared with ICH group(P0.05).Compared with model group,volume density,numerical density of the extract of Leave Ginkgo Biloba treatment group increased notably(P0.05).Conclusion:Leave Ginkgo Biloba extract could protect neurons and promote recovery of the neural function.
