水溶性壳聚糖对阻塞性黄疸大鼠肝纤维化的保护作用及其机制 被引量:2

摘要 目的探讨水溶性壳聚糖对阻塞性黄疸大鼠肝纤维化的保护作用及其机制。方法将72只SD雄性大鼠随机分为对照组(SO组),胆总管结扎+生理盐水组(BDL+NS组),胆总管结扎+水溶性壳聚糖组(BDL+WSC组)。分别于术后7、14和21 d检测肝星状细胞内MDA、SOD。用RT-PCR检测Ⅰ、Ⅲ型胶原及TNF-α mRNA的表达。结果 BDL+NS组各时间点肝星状细胞内MDA含量明显升高,而SOD却明显降低。胆总管结扎7、14 d时,肝星状细胞内MDA含量BDL+WSC组比BDL+NS组低,差异有统计学意义(7 d,P<0.01;14 d,P<0.05);21 d时,MDA含量BDL+NS组和BDL+WSC中都更进一步升高,但两组比较差异无统计学意义(P>0.05)。胆总管结扎7 d、14 d时,肝星状细胞内SOD含量BDL+WSC组比BDL+NS组下降程度明显(7 d,P<0.01;14 d,P<0.05);21d时,两组肝星状细胞内SOD含量都进一步增加,但差异无统计学意义(P>0.05)。与BDL+NS组比较,胆总管结扎7 d、14 d时,BDL+WSC组可以明显减少I、III型胶原及TNF-α mRNA的表达。结论水溶性壳聚糖在大鼠阻塞性黄疸早、中期有保护HSC细胞代谢作用,减轻了阻黄时肝脏的纤维化。 Objective To investigate the effects of water-soluble chitosan on fibrosis of liver in the rat with obstructive jaundice.Methods Seventy-two male SD rats were random divided into groups of sham operation group and bile duct-ligation+normal soldium group(BDL+NS)and bileduct-ligation+water-solubility chitosan group(BDL+WSC).Malondialdehyde(MDA)、 superoxide dismutase(SOD) and the mRNA expression of collagen Ⅰ and Ⅲ were examined on days 7,14,21 after operation.Results MDA contents of liver,s HSC were significantly increased at each time point in BDL+NSgroup,while SOD contents of HSC decreased remarkably On the 7 th,14 th day;MDA content of HSC in BDL+WSC group were more Lower than that in BDL+NS group(7 th day,P0.01;14 th day,P0.05)On the 21st day;MDA content were further increased in BDL+NS group and in BDL+WSC group but there was no difference in their comparisons.On the 7 th,14 th day,SOD contents in BDL+WSC group was significantly higher than that In BDL+NS group(7 th day,P0.01;14th day,P0.05),On the 21 st day,SOD contents in BDL+NS group had no difference with that in BDL+WSC group(P0.05).On the 7 th,14 th day,the mRNA expression of collagen Ⅰ and Ⅲ、TNF-α reduced in BDL+WSC group compared with BDL+NS group.Conclusion WSC have a protective effect on fibrosis of liver in the initial,metaphase stage of obstructive jaundice in the rat.
