中国汉族人群GST1 GSTM1基因多态性与血脂水平及冠心病的相关性研究 被引量:2

Association of glutathione S - transferase T1 and M1 gene polymorphisms with lipoprotein levels and coronary heart disease in the Chinese Han population

摘要 目的通过研究中国汉族人群谷胱甘肽硫转移酶T1、M1(glutathione S—transferase,GSTF1、GSTM1)基因多态性与衄脂水平及冠心病(coronary heart disease,CHD)之间的关系,探讨冠心病的遗传易感性。方法采用病例一对照研究的方法,选取255例冠心病患者和与之年龄、性别相匹配的145例健康人作为对照组,采用聚合酶链式反应(polymerase chain reaction,PCR)方法检测GSTT1、GSTM1基因多态性,同时检测各研究对象的空腹血清总胆固醇(TC)、甘油三酯(TG)、载脂蛋白A(apoA)、载脂蛋白B(apoB)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、脂蛋白a(Lpa)等临床资料。结果冠心病组GSTT1、GSTM1、GSTT1/GSTM1阳性基因型频率低于对照组(P=0.007、P=0.003、P=0.005),GSTT1/GSTM1阴性基因型频率高于对照组(P=0.001)。GSTM1阴性基因组apoB、LDL水平高于GSTM1阳性基因组(P=0.005、P:0.009);GSTF1/GSTM1阴性基因组apoB、LDL水平高于GSTT1/GSTM1阳性基因组(P=0.014、P=0.013)。结论GSTT1和GSTM1阴性基因型可能是中国汉族人群冠心病的遗传易感基因,GSTM1基因多态性可能通过影响血脂代谢而参与冠心病的发病过程。 Objective To investigate the genetic susceptibility to coronary heart disease(CHD) by studying the association of glutathione S - transferase T1 and M1 (GSTT1, GSTM1 ) gene polymorphisms with lipoprotein levels and CHD in the Cilinese Han population. Methods In this case -control study, the GSTT1, GSTMI gene polymorphisms were detected by polymerase chain reaction (PCR) method in 255 CHD patients and age - and sex - frequencies matched 145 controls. Clinical parameters such as fasting serum total cholesterol (TC), triglyceride (TG) , apolipoprotein A (apoA), apolipoproteln B (apoB), high - density lipoprotein (HDL), low - density lipoprotein (LDL), lipoprotein a (Lpa), fasting blood glucose and other clinical data were measured. Results (1)The frequencies of GSTT1, GSTM1, GSTT1/GSTM1 positive genotype were lower in CHD group than in control group (P = 0. 007, 0. 003, 0. 005, respectively) and the frequencies of GSTT1/GSTMI negative genotype were higher in CHD group than in control group (P =0. 001 ). (2)The levels of apoB and LDL were higher in GSTM1 negative genotype group than in GSTM1 positive genotype group ( P = 0. 005, 0. 009, respectively), the indicators were higher in GSTT1/GSTM1 negative genotype group than in GSTT1/GSTM1 positive genotype group in all subjects (P =0. 014, 0. 013, respectively). Conclusion GSTI'I and GSTM1 negative genotypes are genetic susceptibility factors of coronary heart disease in Chinese Han population , GSTM1 gene polymorphisms may be involved in the pathogenesis of coronary heart disease by influencing the lipid metabolism .
