摘要
目的观察转化生长因子β1(TGF-β1)抗体对小鼠肺成纤维细胞增殖及Ⅲ型胶原的作用。方法用正交设计方法,选取不同水平二氧化硅(SiO2)、TGF-β1抗体及其作用的先后顺序和两者作用的时间间隔4个因素,用四甲基偶氮唑盐(MTT)法检测对小鼠肺成纤维细胞增殖的影响;用免疫组化法检测SiO2、TGF-β1抗体对小鼠肺成纤维细胞Ⅲ型胶原表达的影响。结果不同水平SiO2、TGF-β1抗体及其作用的先后顺序对细胞增殖有显著影响(P<0.05),两因素作用的时间间隔对结果影响差异未见有统计学意义;TGF-β1抗体处理组Ⅲ型胶原表达显著低于SiO2处理组(P<0.05)。结论 TGF-β1抗体能降低小鼠肺成纤维细胞增殖及Ⅲ型胶原表达。
Objective To observe the effects of transforming growth factor-β1(TGF-β1) antibody on the mouse lung fibroblast proliferation and type Ⅲ collagen expression.Methods With the orthogonal design method,MTT method was used to study mouse lung fibroblast proliferation at different concentrations of SiO2 and TGF-β1 antibody,functionary order and the time interval between SiO2 and TGF-β1 antibody.The effect of SiO2,TGF-β1 antibody to type Ⅲ collagen expression of the mouse lung fibroblast was detected by immunological method.Results There were significant effect on cell proliferation among different le-vels of SiO2,TGF-β1 antibody and its functionary order(P0.05),while there were no significant differences between different time intervals.Ⅲ collagen expression in TGF-β1 antibody group was significantly lower than that in SiO2 group(P0.05).Conclusion Mouse lung fibroblast proliferation and type Ⅲ collagen expression could be reduced by TGF-β1 antibody.
