摘要
采用分子荧光光度法测定银杏叶中硒的含量,样品用硝酸和高氯酸混酸消解后,用盐酸羟胺将六价硒还原成四价硒,在pH=2.0的条件下与DAN生成荧光物质4,5-苯并苤硒脑,再用环己烷萃取,在激发波长378 nm,发射波长519 nm下测定其荧光强度。方法的线性相关系数r=0.9905,平均加标回收率为98%,该方法用于样品的分析测定,结果良好。
A new method was developed for the determination of selenium by molecular fluorescence spectrometry.The effects of various factors,such as pH value and quantity of 2,3-diaminonaphthalene on fluorescence were discussed.The nitric-perchloric mixed acid was used to digest the sample,and the selenium(VI)was reduced to selenium(IV) by hydroxylamine hydrochloride.The fluorescence was detected in excitation wavelength(378.0 nm) and emission wavelength(519 nm).The linearly dependent coefficient was 0.9905 and the spiking average recovery was 98%.The results show that the method is reliable and sensitive on real samples.
