摘要
目的探讨肉豆蔻提取物对小鼠小胶质BV2细胞的作用机制。方法运用WST-8法检测肉豆蔻提取物对小鼠小胶质BV2细胞生存率的影响,倒置显微镜观察其形态学改变,Western Blot检测脂多糖(LPS)所诱导的小鼠小胶质细胞环氧合酶2(COX-2)的表达。结果肉豆蔻提取物对小鼠小胶质BV2细胞无明显毒性;可抑制谷氨酸对小鼠小胶质BV2细胞的形态学损害;抑制LPS所诱导的小鼠小胶质BV2细胞COX-2的表达。结论肉豆蔻提取物对小鼠小胶质BV2小胶质细胞具有保护作用。
Objective To study the mechanism of myristica fragrans extracts on murine microglial BV2 cells.Methods WST-8 method was used to detect the effect of myristica fragrans extract on viability rate of murine microglial BV2 cells,inverted microscope was employed to observed its morphological changes and Western Blot was conducted to measure the expression of cyclooxygenase 2(COX-2) in murine microglial BV2 cells induced by lipopolysaccharide(LPS).Results Myristica fragrans extracts showed no sign of obvious toxicity on murine microglial BV2 cells,but it could inhibit glutamic acid caused damage to murine microglial BV2 cells and inhibit expression of COX-2 in murine microglial BV2 cells induced by LPS.Conclusion Myristica fragrans extracts show a protective effect on murine microglial BV2 cells.
