摘要 目的探讨熊果酸在体外对急性早幼粒白血病HL60细胞增殖抑制和诱导凋亡作用。方法采用体外细胞培养技术,荧光显微镜观察细胞结构变化、MTT法观察细胞生长抑制作用、流式细胞仪检测细胞凋亡及周期变化。结果 MTT法证实熊果酸对HL60细胞具有增殖抑制和杀伤效应,并呈浓度和时间依赖性。显微镜下可见HL60细胞出现典型凋亡细胞形态学改变。流式细胞仪检测细胞周期阻滞于G0/G1期。结论熊果酸在体外可抑制急性早幼粒白血病HL60细胞增殖,诱导细胞凋亡。 Objective To investigate anti-proliferation and apoptosis effects of Ursolic Acid on HL60 cells.Methods By culturing in vitro and treated with UA,anti-proliferation effects were assessed with methyl thiazolyl tetrazolium(MTT) colorimetric assay and morphology of cells was observed under fluorescence microscope.The flow cytometry was used to detect apoptotic rates and cell cycle distribution.Results MTT showed that anti-proliferation effect of UA on HL60 Cells in a concentration and time dependent manner.Microscope showed that cell presented some morphologic features of apoptosis.The flow cytometry showed that after exposure to drugs cell-cycle was blocked at G0 /G1 phase.Conclusion Ursolic Acid inhibits proliferation of HL60 cell and induces apoptosis.
