摘要
目的探讨CD24在黑色素瘤细胞增殖中的作用。方法 B16F10细胞转染靶向CD24的RNA干扰质粒pSi-CD24,然后用RT-PCR和WesternBlot观察其对细胞CD24表达的影响,用MTT法观察其对细胞增殖的抑制效果。结果在mRNA和蛋白水平均证实,转染pSi-CD24明显抑制B16F10细胞CD24的表达;与对照组相比较,pSi-CD24转染在48h和72h显著抑制细胞增殖速度。结论 CD24具有促进黑色素瘤细胞体外增殖作用,在肿瘤的发生发展中起重要作用。
Objective To study the effect of CD24 on the proliferation ability of melanoma cells.Methods B16F10 cells were transfect with pSi-CD24,a siRNA plasmid target to CD24.Then the expression of CD24 was measured by RT-PCR and Western blot and the cell proliferation was assessed by MTT assay.Results CD24 expression was obviously down-regulated by pSi-CD24,and knockdown CD24 can effectively inhibited the proliferation of melanoma cells.Conclusion CD24 can enhance the proliferation of melanoma cells in vitro,which indicated CD24 play an important role in tumor growth.
