摘要
目的:研究不同浓度肉苁蓉总苷对体外培养大鼠嗅鞘细胞及其分泌神经营养因子水平的影响。方法:采用MTT测定法和流式细胞计测定法检测肉苁蓉总苷对嗅鞘细胞(OECs)增值的影响;用ELISA法测定OECs分泌胶质细胞源性神经营养因子(GDNF)的含量。结果:MTT测定结果显示10μg/ml和100μg/ml浓度肉苁蓉总苷能明显促进OECs增殖;流式细胞计测定结果表明肉苁蓉总苷能促进嗅鞘细胞G1周期的增殖,并能减少嗅鞘细胞凋亡数量;ELISA测定结果表明100μg/ml浓度肉苁蓉总苷能对OECs分泌GDNF有显著的促进作用。结论:肉苁蓉总苷能促进OECs增殖,并能有效促进其GDNF的分泌。
Objective:To explore the effects of different concentration of cistanche total glycosides on promoting olfactory ensheathing cells'(OECs) proliferation and glial-cell-line-derived neurotrophic factor (GDNF) secretion. Method: The promoting effect of cistanche total glycosides was observed by flow cytometry and MTT, while GDNF by ELISA. Result: The 10μg/ml and 100μg/ml concentration of cistanche total glycosides could promote OECs' proliferation;100μg/ml concentration of cistanche total glycosides could promote OECs' GDNF secretion. Conclusion: Cistanche total glycosides could promote OECs' proliferation and GDNF secretion.
