摘要
目的:观察注射氢水对一次性力竭运动大鼠骨骼肌氧化应激损伤的影响。方法:30只健康雄性SD大鼠,按体重配对后随机分成安静对照组(C组)、运动对照组(E组)与运动给氢组(H组)3组,每组10只。C组不运动。H组于运动前按10 ml.kg-1体重剂量腹腔注射氢水,E组注射同体积生理盐水,然后以0坡度、28 m.min-1速度进行一次性力竭跑台运动,力竭运动后3 h取后肢腓肠肌,用Elisa法测定其3-硝基酪氨酸(3-NT)和8-羟基脱氧鸟苷(8-OHdG)浓度,用化学比色法测定MDA、SOD、GSH与TAOC。结果:与C组比较,E组3-NT、MDA与8-OHdG均显著升高(P<0.05,P<0.01,P<0.01),SOD、GSH含量显著降低(P<0.05),TAOC有所降低,但无统计学意义;H组3-NT、SOD、GSH、TAOC与C组比较无显著性差异,MDA显著低于C组(P<0.01),8-OHdG浓度显著高于C组(P<0.05)。H组3-NT与E组比较无明显变化,但MDA、8-OHdG显著降低(P<0.01,P<0.05),SOD、GSH、TAOC显著升高(P<0.01,P<0.01,P<0.05)。结论:氢水能降低骨骼肌自由基代谢水平,提高骨骼肌抗氧化酶活性,增强骨骼肌的总抗氧化能力,对骨骼肌运动性氧化应激损伤具有保护作用。
Objective To study the effects of supplementing hydrogen-rich water on the oxidation stress-induced damage in skeletal muscle after single bout of exhaustive exercise.Methods Thirty healthy SD male rats were equally and randomly divided into:sedentary control group(C),exercise control group(E)and exercise + hydrogen water group(H).Intraperitoneal injection of hydrogen-rich water(10 ml·kg-1)was performed for group H,and the same amount of normal saline was intraperitoneally injected to group E one minute before single bout of exhaustive exercise on a treadmill at the speed of 28 m·min-1 with 0° slope.3 hours after exercise,the gastrocnemius was drawn to determine the density of 3-nitrotyrosine(3-NT)and 8-hydroxy deoxyguanosine(8-OHdG)by Elisa measurement,and MDA,SOD,GSH and TAOC by chemical colorimetry.Results Compared with group C,3-NT,MDA and 8-OHdG of group E significantly elevated(P 0.05,P 0.01,and P 0.01,respectively),while SOD and GSH significantly reduced(P 0.05),however,TAOC reduced insignificantly;MDA was significantly lower(P 0.01)and 8-OHdG was remarkably higher(P 0.05),whereas 3-NT,SOD,GSH and TAOC of group H and C changed significantly.When compared with group E,MDA and 8-OHdG of group H significantly decreased(P 0.01 and P 0.05,respectively),SOD,GSH and TAOC significantly increased(P 0.01,P 0.01,P 0.05,respectively),whereas 3-NT changed insignificantly.Conclusion Hydrogen-rich water could alleviate the metabolic level of free radical,elevate the activity of antioxidase,and thus protect stress-induced damage in skeletal muscle after acute exhaustive exercise.
