摘要
目的以Aβ1-42诱导原代海马神经细胞建立阿尔茨海默病细胞模型。方法体外培养原代海马神经细胞,加入不同浓度Aβ1-42诱导并收获细胞,采用MTT法观察细胞活力,流式细胞术观察细胞凋亡及继发性死亡情况。结果 Aβ1-42作用于神经细胞后细胞活力明显下降,细胞存活率呈浓度依赖性降低;流式结果显示细胞凋亡及继发性死亡呈浓度依赖性增加。结论 Aβ1-42诱导后海马神经细胞存活率下降,细胞凋亡,可作为理想的AD细胞模型。
Objective To set up the Alzheimer′s disease(AD) cell model using hippocampal cells of primary culture induced by Aβ1~42.Methods The hippocampal cells were cultured in vitro and Aβ1~42 was added into the medium in different concentrations and then MTT assay was used to observe the cell vitality and Annexin V/PI double staining to analyze the apoptosis and the secondary death by flow cytometry.Results The cell vitality was decreased significantly after being induced with Aβ1~42 and the apoptosis and the secondary death were increased in a dose-dependent manner.Conclusions The cell vitality is decreased with the apoptosis of the hippocampal cell induced with Aβ1~42,which is as a good AD cell model.
