摘要
目的观察化瘀泄浊方对肾间质纤维化(RIF)大鼠骨形态发生蛋白(BMP)-7的影响。方法选用健康、雌性SD大鼠50只,用单侧输尿管结扎方法复制大鼠RIF模型,将其随机分为假手术组、化瘀泄浊方组(化瘀泄浊方5.7 g.kg-1.d-1经口灌服)、阿魏酸哌嗪分散片组(予阿魏酸哌嗪分散片6.7 mg.kg-1.d-1)、缬沙坦组(予缬沙坦5.3 mg.kg-1.d-1经口灌服)及模型组(予同等量生理盐水经口灌服),每组10只大鼠。行HE、Masson染色观察梗阻侧肾脏组织形态学改变并用免疫组化方法检测观察梗阻侧肾脏BMP-7表达水平。结果模型组及各治疗组与假手术组相比均呈现程度不同的病理损害,梗阻肾间质BMP-7蛋白表达也明显减少(P<0.01)。各治疗组与模型组比较,BMP-7表达明显增加(P<0.01)。治疗各组间比较,化瘀泄浊方组优于阿魏酸哌嗪分散片组(P<0.01),其他各组间均无明显差异(P>0.05)。结论输尿管梗阻后肾组织呈现程度不同的病理损害,肾纤维化面积明显增加,这可能与BMP-7表达减少有关,化瘀泄浊方对其有一定的上调作用。
Objective To observe the effect of Huayuxiezhuofang on BMP-7 in rats with tubulointerstitial fibrosis.Methods The model of renal obstruction was established by unilateral ureteral obstruction(UUO) in rats and then divided into five groups,including sham-operation group,UUOZY group(treated with Chinese medicine of huayuxiezhuo(5.7 g·kg-1·d-1)),UUOA group(treated with ferulaic acid piperazidine dispersible tablet(6.7 mg·kg-1·d-1)),UUOV group(treated with Valsartan(5.3 mg·kg-1·d-1)) and UUO group.The morphological change of kidney tissue and protein expressions of BMP-7 in kidney tissue were observed.Results In UUO group and the different treatment groups there were different degrees of lesions of kidney compared with those of sham-operation group.The expression of BMP-7 in kidney tissue was decreased obviously(P0.05).Compared with UUO group,the expression of BMP7 was increased significantly in treatment groups(P0.05).The comparison between different treatment groups showed that BMP-7 expression in UUOZY group was better than that in UUOA group(P0.05),but there was no significant difference among other treatment groups(P0.05).Conclusions After unilateral ureteral obstruction the different degrees of pathological changes in kidney tissue are seen and the areas of fibrosis is increased obviously,which is related to BMP-7 expression.Chinese medicine of huayuxiezhuo has an up-regulation effect.
