摘要
目的探讨姜黄素是否能够通过下调IκBα的磷酸化而抑制食管鳞癌细胞的增殖并对其细胞周期产生影响。方法 MTT法检测姜黄素作用下食管鳞癌细胞的增殖;Western blot法检测EC9706和Eca109细胞在姜黄素作用下pIκBα和细胞周期蛋白cyclin D1的表达情况。两种细胞中加入姜黄素培养72 h,或联合5-FU培养72 h,流式细胞仪检测细胞周期。结果 EC9706和Eca109细胞的存活率均随着姜黄素浓度的增加逐渐下降;姜黄素作用下EC9706和Eca109细胞中pIκBα和cyclin D1蛋白的表达水平随着时间的延长逐渐下降,且G0/G1期的细胞开始增加,S期的细胞逐渐减少;当姜黄素联合使用5-FU时,G0/G1期的细胞明显增加,S期的细胞明显减少。结论 姜黄素通过下调IκBα及cyclin D1的表达而抑制食管鳞癌细胞的增殖,或许有望成为食管癌治疗中的一个辅助用药。
Objective To detect whether curcumin inhibits NF-κB signaling pathway through suppression IκBα phosphorylation and evaluate the effects on proliferation and cell cycle in EC9706 and Eca109 cell lines.MethodsESCC cells were treated with different concentrations of curcumin and viable cells were determined by MTT.Cytoplasmic extracts or whole cell extracts were examined for the expression of pIκBα and cyclin D1.ESCC cells were treated with curcumin,5-FU or combinations thereof for 72 h.Results Curcumin inhibited ESCC cells growth in a concentration-dependent manner.Curcumin inhibited IκBα phosphorylation and downregulated the expression of cyclin D1 in the two ESCC cell lines in a time-dependent manner.Cyclin D1 was declined in the forepart of curcumin-treated cells.Curcumin-treated cells increased in the G0/G1 phase and decreased in S phase.ConclusionCurcumin inhibits the phosphorylation of IκBα and downregulates the activation of NF-κB signaling pathway,leading to the suppression of proliferation in ESCC cell.
