摘要
目的建立小鼠骨髓抑制模型。方法选用8~10周的♀BALB/c小鼠,体质量20~22 g。每只小鼠腹腔注射1mg的卡铂注射液后立即行6 MV直线加速器照射,照射剂量为每只2 Gy,从而产生骨髓抑制。观察小鼠血常规;骨髓涂片进行HE染色,观察骨髓的变化。结果模型组小鼠有潜在出血倾向,外周血各系细胞下降明显,红细胞计数、血红蛋白含量、血小板计数均在卡铂和放疗联合处理后14 d达到最低点,此后慢慢恢复,处理后28 d可基本恢复至正常水平。相比红细胞计数和血红蛋白含量,血小板计数恢复较为缓慢。外周血白细胞计数在卡铂和放疗联合处理后7 d达到最低点,此后慢慢恢复,至处理后28 d可基本恢复到正常水平。结论腹腔注射卡铂注射液和放疗共同诱导的小鼠骨髓抑制模型重复性好,易于操作,可以作为其他药物研究的基础。
Aim To establish a reliable and practical mice model with myelosuppression.Methods Eight-to ten-week-old female BALB/c mice were prepared.Myelosuppression was induced by intraperitoneal injection of carboplatin at 1.0 mg per mouse,immediately followed by a single dose of total body radiation at 2 Gy.To observe the changes of blood and marrow through blood routine,marrow smears stained by HE.Results In model mice,peripheral blood cells decreased significantly,and red blood cells,hemoglobin and platelets reached their lowest levels on 14th day after the treatment of carboplatin and radiotherapy,and then they restored to normal levels on 28th day after the treatment.Compared to red blood cells and hemoglobin,the recovery of platelets was slow.WBC reached its lowest point on 7th day after the treatment,then slowly recovered to normal level on 28th day.Conclusion The model is reproducible and easy to operate,and it can be used as a tool for studying other drugs.
