摘要 胰岛素样生长因子家簇(IGFs)与肝癌(HCC)发生发展、转移和耐药相关。IGF-ⅠR和IGF-Ⅱ在HCC进展过程中过量表达,IGF-ⅠR与IGF-Ⅱ结合导致自身酪氨酸激酶磷酸化,激活下游信号通路,使肝细胞恶性转化潜力增加,已证实IGF-ⅠR是HCC分子治疗的理想靶点。本文概述了IGF-ⅠR在HCC发生发展过程中表达、作用及所介导的肝癌靶向治疗进展。 Insulin-like growth factors(IGFs) associate with tumorigenesis,metastasis,and drug-resistance of hepatocellular carcinoma(HCC).Both IGF-Ⅰ receptor and IGF-Ⅱ are frequently over expression in HCC progression.Binding of IGF-Ⅱ combined with IGF-ⅠR activates its intrinsic tyrosine kinase activity leading to signaling through cellular pathways increasing the tumorigenic potential of liver.IGF-ⅠR has recently been accepted as a credible treatment target.In recently,more and more evidence suggest molecule targeted IGF-ⅠR also rationally in the treatment of HCC.In this review,we summarize the interaction of IGF-ⅠR with the possible causes of HCC and its potential role in hepato-carcinogenesis as well as current targeting IGF-ⅠR approaches in vitro and/or in vivo.
