生长激素对离体大鼠缺血再灌注心脏泵功能的影响

摘要 目的 应用离体大鼠心脏缺血再灌注模型观察基因重组生长激素( G H)对缺血再灌注心脏泵功能的影响。方法 大鼠随机分为二组,分别每天皮下注射 G H 05 I U/kg( G H 组)或等量生理盐水(对照组),7 d 后进行离体心脏缺血再灌注,分别测定缺血前、后再灌注 10 m in、30 m in 的心脏泵功能指标和冠脉流量的恢复率。结果  G H 组离体心脏缺血前基础水平的左室收缩峰压( L V S P)、收缩期左室压力上升速度峰值(+ dp/dt m ax)、舒张期左室压力下降速度峰值(- dp/dt m ax )和冠脉流量( C F) 与对照组比较呈显著差异( P < 001), 缺血后再灌注10 m in、30 m in 的 L V S P、±dp/dt m ax、 C F 与自身缺血前基础水平比较的恢复百分率明显高于对照组( P <005),而 G H 组心率( H R)的恢复率与对照组比较无显著差异(( P > 005))。结论 提示预先应用 G H 可增强心脏泵功能并改善缺血再灌注心脏泵功能的损伤。 Objective To determ ine w hether grow th horm one ( G H) has effects on pum p function of isolatedm yocardial ischem ia reperfusion heart in rats. Methods G H (0.5 I U/kg s.c. per day) or equal volum e 0.9% saline w ater w ere adm inistered to rats of G H groups and control groups (both n = 10) respectively for7 days. A m odel of acute ischem ia reperfusion injury w as produced in anaesthetized open chest rats. Theindices of left ventricular pum p function, in term s of L V S P, ± dp/dtm ax, H R and C F, w ere determ inedbefore m yocardial ischem ia and during reperfusion (10, 30 m in. respectively). Preischem ic values w ererecorded directly as baseline values for each group and postischem ic values w ere expressed by percent re covery of baseline values. Results It show ed that the baseline values of left ventricular pum p function asw ellas percent recoveries of L V S P, ± dp/dtm ax and C F during reperfusion (10,30 m in) in G H groupw ere significantly higher than those in control group, ( P < 0.01 and P < 0,05, respectively) . The per cent recovery of H R had no significant difference betw een tw o groups ( P > 0.05) . Conclusion G H has ef fects on pum p function of isolated m yocardial ischem ic heart in rats.
