L-精氨酸对大鼠肾缺血再灌注损伤HSP70表达的影响 被引量:1

摘要 目的:探讨左旋精氨酸(L-arg)对大鼠肾缺血再灌注损伤(RIRI)HSP70表达的影响。方法:健康雄性SD大鼠48只,随机分为6组:假手术(Sham)6 h、24 h组;缺血再灌模型(IR)6 h2、4 h组和L-精氨酸(L-arg)6 h2、4 h组。IR组和L-arg组采用夹闭双侧肾动、静脉45 min后松夹再灌的方法制备RIRI模型,L-arg组于夹闭前和再灌前15 min分别尾静脉注射L-arg 0.2 mL(100 mg/kg),于再灌后6 h和24 h测定血清肌酐(Scr)、一氧化氮(NO)及尿NAG酶(UNAG)含量,观察肾组织形态学改变和肾皮质HSP70表达。结果:缺血再灌注后6 h和24 h,大鼠肾功能和肾小管上皮细胞明显受损,Scr和UNAG含量均明显高于假手术组(P<0.01),血清NO及肾皮质细胞HSP70表达明显增加。尾静脉注射L-arg可明显改善RIRI大鼠肾功能,减轻肾损伤,并明显增加血清NO及肾皮质细胞HSP70表达(P<0.05)。结论:L-arg可通过增加体内NO水平,提高肾小管上皮细胞HSP70表达,对大鼠RIRI起到一定的保护作用。 Objective:To investigate the possible role of L-arginine in the pathogenesis of the heat shock protein 70 in renal cortex cells of renal ischemia-reperfusion injury(RIRI) in rats.Methods:48 SD rats were divided into 6 groups randomly:Sham 6 h,24 h groups,IR 6 h,24 h groups and L-arg 6 h,24 h groups.The model of RIRI was induced by bilateral clamping the renal artery and vein for 45 minutes followed by reperfusion.L-arg 100 mg/kg was injected via the tail vein before ischemia and 15 min before reperfusion.Serum creatinine(Scr) and urea N-Acety-P-beta-Glucosaminidase(UNAG) were measured for assessment of renal function.Nitric oxide(NO2-/NO3-) and renal histological changes were tested for determination the degree of renal injury.Flow cytometer was made for estimate the contents of HSP70 in renal cortex cells.Results: Contents of Scr and UNAG were significantly increased in RIRI rats(P0.01),serum NO levels and the expression of HSP70 were both markedly increased in RIRI rats.L-arg might significantly decrease the contents of Scr and UNAG.L-arg also could significantly enhance the expression of serum NO and the HSP70 levels of renal cortex cells consequently and attenuated the renal histological injury in RIRI rats.Conclusion:L-arg has beneficial effects on renal dysfunction in RIRI rats through production NO and enhances the HSP70 expression of the renal cortex cells.
