摘要
目的:研究大麻提取物在体内外对人结肠癌细胞LS174T的生长抑制及诱导凋亡作用。方法:体外培养结肠癌细胞,加以不同浓度(500,250,125,62.5,31.25μg/mL)的大麻提取物培养72 h,用MTT法检测大麻提取物对结肠癌细胞抗增殖的作用;用DNA末端原位标记染(TUNEL)法检测大麻提取物对结肠癌细胞的诱导凋亡作用;建立结肠癌小鼠模型,将实验动物随机分为五组:空白对照组、阳性药物组、低剂量组(25 mg/kg)、中剂量组(50 mg/kg)、高剂量组(100 mg/kg),每组5只小鼠,2周后处死动物测定肿瘤的重量和体积。结果:不同浓度的大麻提取物处理细胞72 h后,结肠癌细胞的抑制率分别为(93.12±2.66)%(、74.56±4.62)%(、62.37±3.77)%(、36.84±6.24)%(、19.16±4.24)%,各组间抑制率相比,差异有统计学意义(P<0.05);用TUNEL法可检测到典型的结肠癌细胞凋亡形态;在结肠癌小鼠模型上给药组肿瘤生长被抑制,其肿瘤体积、重量均较对照组减小,差异具有统计学意义(P<0.05)。结论:大麻提取物在体内外具有抑制结肠癌细胞LS174T增殖及诱导凋亡的作用,且呈剂量依赖性。
Objective:To study the Cannador inhibited proliferation and induced apoptosis of human colon carcinoma cells LS174T in vivo and vitro.Methods:Huamn colon carcinoma cells were incubated with different concentration of Cannador for 24,48 and 72 hours,cells viability was measured by MTT assay;Morphological changes of apoptotic cells was detected by TUNEL staining;Nude mouse model with colon carcinoma were established and separated five groups: blank control group,medicine control group,low-dose group(25 mg/kg),meta-dose group(50 mg/kg),high-dose group(100 mg/kg).Two weeks later nude mouse were put to death and then tested the weight and volume of tumour.Results:Cell inhibition ratio with different concentration of Cannador for 24,48 and 72 hours were(93.12±2.66)%、(74.56±4.62)%、(62.37±3.77)%、(36.84±6.24)%、(19.16±4.24)% respectively.There was a significant distinction between groups(P0.05);Typical morphological features of apoptotic cells were detected by TUNEL staining.The growth of tumor in nude mouse was inhibited by different concertration of Cannador,and the weight and volume of tumor were smaller than control group(P0.05).Conclusion:Cannador can inhibit effectivly proliferation and induce apoptosis of huamn colon carcinoma cells LS174T in vivo and vitro and dosage-time-dependently.
