超高效液相色谱-飞行时间质谱联用分析柏芩清火颗粒中的化学成分(Ⅱ)

摘要 目的分析柏芩清火颗粒中的化学成分。方法采用超高效液相色谱-飞行时间质谱联用(UPLC-TOF-MS)方法分析。色谱柱为ACQUITY UPLC BEH C18柱(2.1 mm×100 mm,1.7μm),流动相为0.1%甲酸溶液(A)-0.1%甲酸乙腈溶液(B),流速为0.35 mL.min-1。ESI-MS检测采用正离子模式,以检测生物碱等成分。通过与组方的单味药对比判断色谱峰的来源药材,然后通过与来源药材化学成分进行精确分子量的对比进行化合物的鉴定。结果对柏芩清火颗粒中来源于黄柏、黄芩、黄连等5味药材中的13个化合物进行了鉴定,包括黄柏碱、小檗碱等3个生物碱,黄芩苷、汉黄芩苷等6个黄酮类成分,仙鹤草内酯和关黄柏内酯A 2个内酯类成分,1个三萜皂苷类成分甘草酸及1个酚酸类成分3-O-阿魏酰奎宁酸。结论在中药复杂成分的研究中,质谱的不同检测模式可以互相补充,使复方制剂的成分得到更完整地体现。 Objective To qualitatively analyze the chemical constituents of Baiqin Qinghuo Keli.Methods UPLC-TOF-MS was used.ACQUITY UPLC BEH C18 column(2.1 mm × 100 mm,1.7 μm) was used with 0.1% formic acid solution(A) and 0.1% formic acid in acetonitrile(B) at 0.35 mL·min-1 as the mobile phase.ESI-MS data were acquired in a positive mode.Major constituents in the preparation were characterized by matching their accurate mass with that of the constituents of source herbs.Results A total of 13 compounds were derived from 5 herbs:phellodendrine,baicalin,and berberine were indentified in the preparation,including 3 alkaloids such as phellodendrine and berberine,6 flavonoids such as baicalin,wogonoside,2 lactones,namely amurenlactone A and agrimonolide,1 triterpene saponin glycyrrhizic acid and 1 phonolic acid 3-O-feruloylquinic acid.Conclusion UPLC-TOF-MS analysis with different detection modes can provide supplementary information on the complex chemical constituents of traditional Chinese medicine,which can be used in combination to profile the components of traditional Chinese formulae more efficiently.
