肺癌线粒体HV1基因突变的初步研究 被引量:1

摘要 目的探讨线粒体HV1基因突变与肺癌的相关性,同时评价痰液中检测HV1基因突变的诊断学意义。方法从38例肺癌患者的蜡块组织及痰液中提取DNA,同时以31例非肺癌(炎症或结核)患者的蜡块组织作为对照,采用聚合酶链反应产物单链构象多态性(PCR-SSCP)方法分析HV1基因突变。结果 38例肺癌患者的蜡块组织中发现21例存在异常迁移带,其中中央型肺癌16例,周围型肺癌5例;31例对照组织中发现8例存在异常迁移带;组织中存在异常迁移带的21例肺癌患者痰液中发现14例存在与组织相同的异常迁移带,中央型肺癌13例,周围型肺癌1例;16例中央型肺癌患者按肿瘤细胞分化程度分高、中、低分化3组,3组患者痰液中HV1基因突变阳性率分别为33.3%(1/3)、80%(4/5)和100%(8/8)。结论线粒体HV1基因突变与肺癌之间存在一定的相关性;肺癌患者痰液中HV1基因突变阳性率达66.7%(14/21),同时中央型肺癌患者痰液中HV1基因突变阳性率明显高于周围型,并且与肿瘤细胞的恶性程度呈正相关。
