枸杞多糖对THp-1单核细胞源性巨噬细胞内质网应激的影响 被引量:2

The Effect of Lycium Barbarum Polysaccharides on Expression of Endoplasmic Reticulum Stress in THP-1 Monocyte-derived Macrophages

摘要 目的研究枸杞多糖(LBP)对THP-1单核细胞源性巨噬细胞内内质网应激反应蛋白及氧化低密度脂蛋白(ox-LDL)含量变化的影响。方法培养THP-1单核细胞,加入佛波酯Qhorb0112-myristatc13-acetat,PMA)诱导分化成THP-1巨噬细胞,随后,加入不同浓度的LBP(207g/ml、507g/ml、1007g/ml、2007g/m1)孵育24h后收集细胞。用酶联免疫吸附法(ELIsA)检测细胞上清液中OX-LDL及内质网应激反应蛋白(Grp78,CHOP,Xbp-1)的含量变化。结果不同浓度的LBP孵育THP-1巨噬细胞,细胞中的OX-LDL与内质网应激反应蛋白的含量均降低。结论LBP可以降低OX-LDL及内质网应激反应蛋白的含量,对预防动脉粥样硬化的发生发展起到一定的作用。 Objective To study the effect of Lycium barbarum polysaccharides on expression of endoplasmic reticulum stress in macrophages derived from THP-1 monocytes.Methods THP-1 monocytes were induced into THP-1 macrophages by culturing with PMA.After exposure of the cultured THP-1 monocytes to LBP at increasing concentrations(0,20,50,100 and 200?g/ml) for 24 h.ELISA was used to determine the expression of Grp78,CHOP,Xbp-1 and ox-LDL in each group.Results THP-1 monocyte-derived macrophages were treated with different concentrations of LBP,we found that the expression of ox-LDL and Grp78,CHOP,Xbp-1 were decreased.Conclusion LBP may down-regulate the expression of Grp78,CHOP,Xbp-1 and ox-LDL.
