摘要
在单因素试验的基础上,采用Box-Behnken设计对超声辅助提取大叶金花草总黄酮工艺中的液料比、提取时间和超声功率3因子的最优化组合进行了定量研究,建立并分析了各因子与得率关系的数学模型。结果表明:最佳的工艺条件为液料比9.9mL/g、超声温度60.1℃和超声时间51.8m in。经试验验证,在此条件下总黄酮得率为6.50%,与理论计算值6.44%基本一致。说明回归模型能较好地预测大叶金花草总黄酮的提取得率。在3个因素中,液料比和超声时间对总黄酮得率的影响显著,超声温度影响对总黄酮得率的不显著。超声辅助提取大叶金花草总黄酮方法简单可行,是一种大叶金花草总黄酮提取的较好方法。
The influences of ethanol concentration, ultrasonic temperature and ultrasonic time on the yield of total flavonoids from Stenoloma chusana ( L. ) Ching with the ultrasonic -assisted extraction technique were studied. Box - Behnken experimental design was applied based on the single factor experiments. One mathematical model was established, response surface methodology was used to study the relationship between the factors and the total flavonoid ex- traction yield. Based on the canonical analysis, the optimum extraction conditions were obtained as follows: solvent to material ratio 9. 9mL/g, ultrasonic temperature 60. 1℃ and ultrasonic time 51.8min. Under the optimized extraction conditions, the total flavonoid extraction yield was 6. 50% , which was close to the estimated value of 6.44% obtained by regression model. Therefore, this model can be used to well predict the experiment results. Among three factors, solvent to material ratio and ultrasonic itime had significant effects on total flavonoid extraction yield, while ultrasonic temperature had remarkable effect as well. This method was simple, efficient and suitable for extracting the total flavonoids from Stenoloma chusana (L.) Ching.
