响应面法优化植物乳杆菌素的双水相萃取条件 被引量:6

Optimization of the Extraction Condition for Plantaricin in Aqueous Two-phase Systems by Response Surface Analysis

摘要 采用聚乙二醇/硫酸铵双水相体系研究发酵上清中植物乳杆菌素的萃取条件。通过单因素试验考察不同PEG分子质量、PEG浓度、硫酸铵浓度、离子强度等对细菌素纯化系数及萃取率的影响。在此基础上,用响应面法对系统进行优化。试验结果表明,植物乳杆菌素萃取的最佳条件为PEG4000-硫酸铵体系,PEG18%,硫酸铵20%、氯化钠1%,萃取pH3.75。 Aqueous two-phase system composed of PEG/(NH4)2SO4 was used to extract plantaricin from cell-free supernatant in this paper.The effects of different PEG molecular weight,PEG concentration,(NH4)2SO4 concentration,NaCl addition etc.on the Purification coefficient and extraction rate of bacteriocin were investigated by single factor test.Subsequently,the response surface methodology was employed to optimize this system.The results showed that the optimum conditions were as follows:the mass composition in two-phase aqueous system 18% PEG4000,20% (NH4)2SO4,1% NaCl,pH 3.75.
