摘要
探讨醋酸铅对体外培养的大鼠肾小管上皮细胞系HBZY-1的细胞毒性效应。用不同浓度的醋酸铅(30、40、50μmol/L)作用HBZY-1细胞24h、48h和72h,采用CellTiter-Glo^(?)萤光细胞活性检测盒测定细胞存活情况;用流式细胞仪检测细胞凋亡率。结果显示经30、40、50μmol/L醋酸铅处理HBZY-1细胞72h后,细胞活性明显降低(P<0.05);流式细胞仪检测结果表明其细胞凋亡率明显升高。醋酸铅可损伤肾小管上皮细胞HBZY-1,促进其凋亡及影响肾脏。
To explore the toxic effect of lead acetate on rat ren 1 tubular epithelial cell HBZY-1. HBZY-1 cells were treated with different concentrations of lead acetate(30,40 and 50 p, mol/L) for 24 h,48 h and 72 h. After 48 hours,using CellTiter-GloTM Luminescent Cell Viability Assay determined cell activity; the apoptosis rate was measured by flow eytometry. The results shows that cell activity decreased significantly( P 〈 0.05 ) by 30,40 and 50 μmol/L lead acetate treatment HBZY-I cells after 72 h; Flow cytometry results showed that the cell apoptosis rate was higher in the treatment group than in the control group. Lead acetate can remarkably inhibit rat renal tubular epithelial HBZY-I cell growth, promote apoptosis and affect the kidney.
