L-氨基酸氧化酶对人肺腺癌A549细胞增殖、凋亡的影响研究 被引量:2

摘要 目的:探讨L-氨基酸氧化酶(LAAO)对人肺腺癌A549细胞增殖、凋亡的影响,为临床上寻找新的肺癌治疗方法提供参考。方法:将A549细胞制成浓度为5×104个/mL的细胞悬液,根据LAAO干预浓度的不同,将其分为4组:2.41、4.82、9.64、19.28mg·mL-1干预组,各组均干预24、48、72h。镜下观察各组细胞形态学改变,采用MTT法检测细胞增殖抑制率,原位末端标记法检测细胞凋亡指数,另设立阴性对照组进行比较。结果:LAAO对A549细胞增殖的抑制率随时间延长和浓度增加而增加(P<0.05),凋亡指数随时间延长和浓度增加而增加(P<0.05)。结论:LAAO可抑制A549细胞增殖,诱导其凋亡,且呈时间、剂量依赖性。 OBJECTIVE: To investigate the effects of L-amino acid oxidas (LAAO) on the proliferation and apoptosis of human lung cancer cell A549,and to provide a new therapeutic option for lung cancer.METHODS: Cell A549 suspension (5×104/mL) was prepared,then divided in to 4 groups according to the concentration of LAAO: 2.41,4.82,9.64 and 19.28 mg·mL-1 intervention groups.All the groups were treated for 24,48 and 72 h.Cellular morphology changes were observed under microscope.The proliferation inhibitory rate was evaluated by MTT assay.The cell apoptosis index was determined by TUNEL method,and compared with negative control group.RESULTS: LAAO could inhibit the proliferation of A549 cells in a dose-time-dependent manner (P0.05).With the increase of LAAO concentration and the duration of administration,the cell apoptosis index was significantly increased (P0.05).CONCLUSION: LAAO can inhibit the proliferation and induce apoptosis of A549 cells in dose-timedependent manner.
