蛋白芯片技术检测抗ENA抗体的临床应用

Clinical application of protein chip in detection of anti-extractable nuclear antigen antibody

摘要 目的评价蛋白芯片技术检测临床标本中抗可提取核抗原(ENA)抗体的应用价值。方法以免疫印迹法(immunoblot,IB)为参比方法,蛋白芯片法(Protein chip)为待评方法,对62份标本的抗ENA抗体进行平行测定,并对两种方法检测结果进行统计学分析。结果经配对χ2检验,P>0.05,两种方法检测结果差异无统计学意义,且两种方法的阳性符合率为91.43%,阴性符合率均为92.59%,总符合率为91.94%,Kappa值=0.84。结论与传统的免疫印迹分析法检测抗ENA抗体相比,蛋白芯片法具有同等的敏感性和特异性,由于操作更简便,判定方式更客观,因此应用蛋白芯片检测抗ENA抗体在临床实践中有很好的应用前景。 Aim To assess the value of protein chip in detection of anti-extractable nuclear antigen antibody. Methods Anti-extractable nuclear antigen antibody in 62 serun samples were detected by immunoblot and protein chip. Results No statistically significantdifferences in detection of anti-extractable nuclear antigen antibody were observed between two methods. The positive coincidence,negative coincidence ,and the total coincidence were 91.43% ,92.59% and 91.94% respectively. The Kappa value is 0.84. Conclusion Protein chip and immunoblot methods had a good consistency in detecting anti-extractable nuclear antigen antibody. Protein chip showed good prospective in detection of anti-extractable nuclear antigen antibody.
